Ultrastructural Changes in the Diaphragm of Aged Emphysema- tous Hamsters

چکیده
In emphysematous hamsters the diaphragm is continuously exposed to an increased workload. Previous lightmicroscopical observations showed only minor changes in relatively young emphysematous hamsters. However, ageing affects the regenerating capacity of muscles. We hypothesised that the diaphragm of aged emphysematous hamsters shows an increase of ultrastructural alterations in comparison to age-matched normal hamsters. This could contribute to a reduced force-generating capacity, which occurs in emphysematous hamsters. After intratracheally instillation of elastase or saline the animals were sacrificed at the age of 15-16 months for in vitro measurements of isometric contractile properties of the diaphragm. The hamsters were perfused in situ with glutaraldehyde for electronmicroscopical investigations. In emphysematous hamsters, twitch and tetanic forces were reduced by ~28% and ~14%, respectively (P<0.001). Compared to control hamsters the diaphragm showed distinct muscle fibre damage (Z-line dislocation, disarray of myofibrils, misaligned sarcomeres, focal degeneration, focal necrosis and segmental necrosis) and more signs of sub-optimal regeneration (fibre splitting and forking) and abortive regeneration. In conclusion, the diaphragm of aged emphysematous hamsters exhibits increased ultrastructural abnormalities, which are likely to contribute to a reduced force generating capacity.

Myosin and actin filament lengths in diaphragms from emphysematous hamsters.
In vitro studies of the diaphragm from emphysematous animals have, in some instances, shown an alteration in its sarcomere length-tension relationship and a decreased maximal specific tension. To our knowledge, it has never been determined whether such functional changes may be indicative of ultrastructural adaptations, e.g., changes in filament lengths and thus cross-bridge number. To address ...
متن کاملEffects of emphysema on diaphragm blood flow during exercise.
Chronic hyperinflation of the lung in emphysema displaces the diaphragm caudally, thereby placing it in a mechanically disadvantageous position and contributing to the increased work of breathing. We tested the hypothesis that total and regional diaphragm blood flows are increased in emphysema, presumably reflecting an increased diaphragm energetic demand. Male Syrian Golden hamsters were rando...

This study was designed to investigate whether the administration of the anabolic steroid nandrolone decanoate is able to antagonize the loss in diaphragm function induced by long-term administration of a low-dose of methylprednisolone in emphysematous hamsters. Normal and emphysematous male hamsters were randomized to receive either saline or methylprednisolone 0.2 mg x kg(-1) x day(-1) for 9 ...
متن کاملProteasome inhibition improves diaphragm function in an animal model for COPD.
Diaphragm muscle weakness in patients with chronic obstructive pulmonary disease (COPD) is associated with increased morbidity and mortality. Recent studies indicate that increased contractile protein degradation by the proteasome contributes to diaphragm weakness in patients with COPD. The aim of the present study was to investigate the effect of proteasome inhibition on diaphragm function and...
متن کاملEffects of emphysema and training on glutathione oxidation in the hamster diaphragm.
Loading of skeletal muscles is associated with increased generation of oxidants, which in turn may impair muscle contractility. We investigated whether the load on the hamster diaphragm imposed by pulmonary emphysema induces oxidative stress, as indicated by glutathione oxidation, and whether the degree of glutathione oxidation is correlated with contractility of the diaphragm. In addition, the...
